    Conscience in Context A study of the nature of conscience relating to its historical development and existential environment

    The topic of conscience has fascinated me for a number of years. In the course of my pastoral work in different parishes, and chaplaincy work with young people and those in hospital, whether for mental or physical reasons, I have encountered countless people who struggled with moral dilemmas or who felt crushed by guilt owing to the gap between their practice and their knowledge of what they were called to do. I have also met individuals who seemed to be unaware that, despite the gravity of the action, what they were doing was in any way wrong. Whether mentioned or left implicit, the conscience of each of these individuals played a vital role in the decision to choose one course of action over another, in judging a completed action to have been right or wrong, or even in exhibiting a state of perplexed uncertainty as to what should be done next. Thinking over these different problems led me to reflect further on the question of erroneous conscience. Could an action that was considered to be wrong by others (particularly by the Magisterium of the Church) be good, virtuous or meritorious if the individual believed it to be so? Do we live in parallel moral universes, where the person ultimately defines what is moral solely by belief or conviction, or do we have access to a ground of universal truth, rooted in our created nature, as gifted by God? Therefore, should the pastor leave the individual in blissful ignorance, or should he try to deepen moral understanding or help develop the moral capacities of the people he encounters? Consideration of questions such as these led me to investigate the possibility of making conscience the subject of doctoral research. However, the resultant thesis is not a study of pastoral problems, in the style of a manualistic analysis of cases of conscience. Rather the study is at the level of fundamental moral theology, presented in the hope that a deepened awareness of the nature and function of conscience will shape my future pastoral activity, and in the hope that it might help others, too, in their understanding of this core notion of morality

    The large NcN_c limit of four-point functions in N=4 super-Yang-Mills theory from anti-de Sitter Supergravity

    We compute the imaginary part of scalar four-point functions in the AdS/CFT correspondence relevant to N=4 super Yang-Mills theory. Unitarity of the AdS supergravity demands that the imaginary parts of the correlation functions factorize into products of lower-point functions. We include the exchange diagrams for scalars as well as gravitons and find explicit expressions for the imaginary parts of these correlators. In momentum space these expressions contain only rational functions and logarithms of the kinematic invariants, in such a manner that the correlator is not a free-field result. The simplicity of these results, however, indicate the possibility of additional symmetry structures in N=4 super Yang-Mills theory in the large NcN_c limit at strong effective coupling.     A feed-forward measurement scheme for periodic noise suppression in atomic magnetometry

    We present an unshielded, double-resonance magnetometer in which we have implemented a feed-forward measurement scheme in order to suppress periodic magnetic noise arising from, and correlated with, the mains electricity alternating current line. The technique described here uses a single sensor to track ambient periodic noise and feed forward to suppress it in a subsequent measurement. This feed forward technique has shown significant noise suppression of electrical mains-noise features of up to 22 dB under the fundamental peak at 50 Hz, 3 dB at the first harmonic (100 Hz), and 21 dB at the second harmonic (150 Hz). This technique is software based, requires no additional hardware, and is easy to implement in an existing magnetometer

    Multi-omics studies demonstrate Toxoplasma gondii-induced metabolic reprogramming of murine dendritic cells

    Toxoplasma gondii is capable of actively invading almost any mammalian cell type including phagocytes. Early events in phagocytic cells such as dendritic cells are not only key to establishing parasite infection, but conversely play a pivotal role in initiating host immunity. It is now recognized that in addition to changes in canonical immune markers and mediators, alteration in metabolism occurs upon activation of phagocytic cells. These metabolic changes are important for supporting the developing immune response, but can affect the availability of nutrients for intracellular pathogens including T. gondii. However, the interaction of T. gondii with these cells and particularly how infection changes their metabolism has not been extensively investigated. Herein, we use a multi-omics approach comprising transcriptomics and metabolomics validated with functional assays to better understand early events in these cells following infection. Analysis of the transcriptome of T. gondii infected bone marrow derived dendritic cells (BMDCs) revealed significant alterations in transcripts associated with cellular metabolism, activation of T cells, inflammation mediated chemokine and cytokine signaling pathways. Multivariant analysis of metabolomic data sets acquired through non-targeted liquid chromatography mass spectroscopy (LCMS) identified metabolites associated with glycolysis, the TCA cycle, oxidative phosphorylation and arginine metabolism as major discriminants between control uninfected and T. gondii infected cells. Consistent with these observations, glucose uptake and lactate dehydrogenase activity were upregulated in T. gondii infected BMDC cultures compared with control BMDCs. Conversely, BMDC mitochondrial membrane potential was reduced in T. gondii-infected cells relative to mitochondria of control BMDCs. These changes to energy metabolism, similar to what has been described following LPS stimulation of BMDCs and macrophages are often termed the Warburg effect. This metabolic reprogramming of cells has been suggested to be an important adaption that provides energy and precursors to facilitate phagocytosis, antigen processing and cytokine production. Other changes to BMDC metabolism are evident following T. gondii infection and include upregulation of arginine degradation concomitant with increased arginase-1 activity and ornithine and proline production. As T. gondii is an arginine auxotroph the resultant reduced cellular arginine levels are likely to curtail parasite multiplication. These results highlight the complex interplay of BMDCs and parasite metabolism within the developing immune response and the consequences for adaptive immunity and pathogen clearance
